TRAF3 regulates IL-6 receptor signaling in T cells through the phosphatase PTPN22

چکیده

Abstract IL-6 is an important signal three cytokine for T helper 17 (Th17) and follicular (Tfh) differentiation. Previous work has suggested a possible role TRAF3 in the function and/or differentiation of these two subsets, but no direct studies have yet been performed. In this study, we examine IL-6R signaling cells. regulates IL-2R type I interferon receptor cells by altering recruitment phosphatases PTPN2 PTPN22, respectively. B cell curtails recruiting thus hypothesized that through PTPN22-dependent mechanism. We investigated using TRAF3-deficient human lymphoma line from cell-specific knockout mouse (T-Traf3−/−). found reduced IL-6-induced STAT1 STAT3 activation are both required full Tfh cells, whereas antagonizes Th17 Interestingly, vitrodifferentiation naïve CD4 to was not affected deficiency. To determine mechanism which promotes response signaling, treated WT Traf3−/−T with presence PTPN22 inhibitor or vehicle control. inhibition restored STAT3, did restore STAT1, suggesting different mechanisms regulation. Together data implicate regulation partly manner, lending additional level understanding Supported NIH R01 AI123107, AI162656, T32 AI007260

Soluble Interleukin-6 (IL-6) Receptor With IL-6 Stimulates Megakaryopoiesis From Human CD341 Cells Through Glycoprotein (gp)130 Signaling

We have recently shown that stimulation of glycoprotein (gp) 130, the membrane-anchored signal transducing receptor component of IL-6, by a complex of human soluble interleukin-6 receptor (sIL-6R) and IL-6 (sIL-6R/IL-6), potently stimulates the ex vivo expansion as well as erythropoiesis of human stem/progenitor cells in the presence of stem cell factor (SCF). Here we show that sIL-6R dose-depe...

TRAF3 Regulates Homeostasis of CD8+ Central Memory T Cells

Our laboratory reported previously that TNF receptor associated factor 3 (TRAF3) is a positive regulator of TCR signaling and T cell function. In the current study, we present new findings that reveal differential roles for TRAF3 in the regulation of CD4+ and CD8(+) T cells. In response to TCR stimulation in vitro, TRAF3 has greater impact in CD4(+) T cells than in CD8+ T cells. However, T cell...

DOCK8 regulates fitness and function of regulatory T cells through modulation of IL-2 signaling.

Foxp3+ Tregs possess potent immunosuppressive activity, which is critical for maintaining immune homeostasis and self-tolerance. Defects in Treg development or function result in inadvertent immune activation and autoimmunity. Despite recent advances in Treg biology, we still do not completely understand the molecular and cellular mechanisms governing the development and suppressive function of...
